Caspase-9也称ICE-LAP6或Mch6,可与细胞色素c和Apaf1形成复合物被激活,并进一步激活细胞凋亡的最关键酶caspase-3,从而触发凋亡级联反应。因此,Caspase-9是细胞凋亡信号转导过程中重要的上游caspase。
细胞凋亡属于程序化细胞死亡,可见于各种器官和细胞,在正常发育、生理、病理过程中对细胞和器官的稳态具有十分重要的调节作用。Caspase (Cysteine-requiring Aspartate Protease)是参与细胞凋亡过程的蛋白酶家族,包含10多个成员。Caspase-9也称ICE-LAP6或Mch6,可与细胞色素c和Apaf1形成复合物被激活,并进一步激活细胞凋亡的最关键酶caspase-3,从而触发凋亡级联反应。因此,Caspase-9是细胞凋亡信号转导过程中重要的上游caspase。其激活可以通过磷酸化进行调控。试剂盒测定原理基于Caspase-9特异水解其多肽底物Ac-LEHD-pNA (N-acetyl-Leu-Glu-His-Asp-p-nitroanilide),释放出游离的硝基苯胺pNA,后者呈黄色在405 nm具有*吸收峰,用可见光分光光度比色方法测定。其吸光度值对应于Caspase-9的水解活性。反应体系100微升,试剂盒提供的试剂,除标准曲线外可进行50或100次样品测定。适用:测定哺乳动物组织、细胞caspase-9活性。
组成与储存:
1. 裂解缓冲液: 20 ml ,4 oC

3. 2 mM Ac-LEHD-pNA底物: 0.5 ml,-20 oC避光
4. 10 mM pNA标准品: 0.5 ml,-20 oC避光
以上标出的量为100次检测,50次检测的试剂量减半。试剂常温运输。到达后按要求储存,1年内稳定。
所需仪器:
酶标仪与96孔酶标板;或可见光分光光度计配100μl容积的石英比色杯。
工作波长:*吸收波长405 nm,如不具备也可在400~450 nm范围内测定。
